Given how the cells could possibly be arranged, it is quite likely that not all of them will be exposed to the detergent. Further, detergents may not be capable of degrading plasma membranes completely at appropriate concentrations - or if a detergent such as SDS is added to a culture of cells at very high concentration, it will probably denature all protein in the cell, making it much harder to study.
